E-mail: [email protected] Throughout the long history of the bacteriophage T4 research, the phenomenon of lysis inhibition (LIN) became an exclusive point of interest of T4 physiology. An ability to control the timing of cell lysis in response to the availability of bacterial hosts in the environment is considered to contribute significantly to the prevalence of T-even phages in nature. In this study, we investigated lysis inhibition in the significantly diverged pseudo T-even bacteriophage RB49. Although the phage RB49 possesses a highly diverged homologue of the T4 major lysis inhibition gene rI, the Escherichia coli BE and K-12 cells infected with RB49 do not display lysis inhibition. In addition, we demonstrated that secondary infection by RB49 phage induces the lysis inhibition of T4-infected cells. The evidence suggests that phylogenetically distant T4-type phages have elements that constitute a signal of the LIN induction.

Characterization of extended-host-range pseudo-T-even bacteriophage Kpp95 isolated on Klebsiella pneumoniae.

Kpp95, isolated on Klebsiella pneumoniae, is a bacteriophage with the morphology of T4-type phages and is capable of rapid lysis of host cells. Its double-stranded genomic DNA (ca. 175 kb, estimated by pulsed-field gel electrophoresis) can be cut only by restriction endonucleases with a cleavage site flanked either by A and T or by T, as tested, suggesting that it contains the modified derivati...

The sequences and activities of RegB endoribonucleases of T4-related bacteriophages.

The RegB endoribonuclease encoded by bacteriophage T4 is a unique sequence-specific nuclease that cleaves in the middle of GGAG or, in a few cases, GGAU tetranucleotides, preferentially those found in the Shine-Dalgarno regions of early phage mRNAs. In this study, we examined the primary structures and functional properties of RegB ribonucleases encoded by T4-related bacteriophages. We show tha...

Escherichia coli Rho factor is involved in lysis of bacteriophage T4-infected cells.

A Rid (Rho interaction deficient) phenotype of bacteriophage T4 mutants was defined by cold-sensitive restriction (lack of plaque formation) on rho+ hosts carrying additional polar mutations in unrelated genes, coupled to suppression (plaque formation) in otherwise isogenic strains carrying either a polarity-suppressing rho or a multicopy plasmid expressing the rho+ allele. This suggests that t...

The roles of the bacteriophage T4 r genes in lysis inhibition and fine-structure genetics: a new perspective.

Seldom has the study of a set of genes contributed more to our understanding of molecular genetics than has the characterization of the rapid-lysis genes of bacteriophage T4. For example, T4 rII mutants were used to define gene structure and mutagen effects at the molecular level and to help unravel the genetic code. The large-plaque morphology of these mutants reflects a block in expressing ly...

Lysis of lysis-inhibited bacteriophage T4-infected cells.

T4 bacteriophage (phage)-infected cells show a marked increase in latent-period length, called lysis inhibition, upon adsorption of additional T4 phages (secondary adsorption). Lysis inhibition is a complex phenotype requiring the activity of at least six T4 genes. Two basic mysteries surround our understanding of the expression of lysis inhibition: (i) the mechanism of initiation (i.e., how se...
